Association of interleukin -6 Polymorphism and mRNA expression in cerebral palsy – a Pilot study

Abstract


Background - Interleukin -6(IL-6) gene and IL-6 protein levels have been linked with increased risk of cerebral palsy(CP). Within the IL-6 gene, the C allele at rs1800795 has  been implicated. Objectives - To find out IL-6 polymorphism rs 1800795 in CP and to study mRNA expression of IL-6 among study group. Methods: Children with CP were enrolled. Clinical and demographic data of the patients has been recorded. Genomic DNA was extracted from peripheral blood lymphocytes. Genotyping was performed using PCR-RFLP technique. IL-6 -174 G/C gene polymorphism was detected by PCR with subsequent restriction analysis of PCR products. The identified genotypes were according to the presence or absence of the enzyme restriction sites i.e. CC, CG, GG. For expression study of IL-6 real-time quantitative reverse transcriptase PCR was used. IL-6 protein level in serum was measured by ELISA . Results: Subjects were 65 CP children and 65 age and sex matched controls. Mean age was 64±38 months (44 males, 21 females) . Polymorphism was not statistically different in CP and controls (CC, CG, GG were  in 45,18, 2 in CP and 39, 24,2 in controls respectively ,p=0.798). IL-6 levels were 138.17±193.8 pg/ml in CP, 120.89±141.24 in controls(p=0.330). IL -6 mRNA level were 8.48±1.1 in CP and 8.64±2.4 in controls ( p=0.692). Sex, presence of birth asphyxia, microcephaly, type of CP had no significant difference in polymorphism (P>0.05). Conclusion : IL-6 polymorphism has no significant role in CP, but needs to be investigated further.
